Intra-articular injection of human synovium-derived mesenchymal stem cells in beagles with surgery-induced osteoarthritis

BackgroundRecently, cell-based tissue engineering approaches using mesenchymal stem cells (MSCs) have been used to treat osteoarthritis (OA). However, the efficacy of human synovium-derived MSCs (hSD-MSCs) has not yet tested in a canine model OA. The purpose this study was investigate therapeutic effects intra-articular hSD-MSC injections OA model.MethodsSixty beagles underwent surgical manipulation induce and received injection 4 weeks after surgery. dogs were divided into five groups (n = 12) according material: G1, sham group; G2, control group injected with phosphate-buffered saline; G3, G4, G5, experimental different dosages (G3, 2.4 × 106 cells; 4.8 9.6 cells). Magnetic resonance imaging (MRI) histopathological immunohistochemical examinations performed 6 24 injection.ResultsMRI revealed significant improvements synovitis hSD-MSC-injected (G3–G5). Histopathologic analyses showed that cartilage structure proteoglycan staining also significantly improved these (G3–G5) further weeks. Immunohistochemical analysis differences levels collagen types I II between hSD-injected (G3–G5), indicating similar extracellular matrix (ECM) composition naïve articular cartilage.ConclusionOur demonstrated for first time ameliorates progression by restoring cartilage, promoting ECM synthesis, inhibiting inflammatory response.
